rnf-5 : unc-95
The pattern of UNC-95::GFP (translational fusion construct) expression was analyzed when RNF-5 was overexpressed under a heat shock promoter. When the wild-type form of RNF-5 was overexpressed in adult animals, a marked decrease in the intensity of GFP labeling was detected after heat shock, especially in the muscle cell body, as compared with control animals (n > 100). However, upon overexpression of RNF-5, which harbors a mutation within the RING finger domain, there was a less pronounced effect on the GFP levels. These findings suggest that RNF-5 affects the stability of UNC-95 and requires an intact RING domain for this activity.
