Removing one copy of the daf-16 gene, in rle-1;daf-16/+ animals, partially suppressed the rle-1 mutant lifespan extension. However, when compared to N2 (or daf-16/+)animals, rle-1/;daf-16/+ worms still live slightly longer.
skn-1(RNAi); daf-16(mgDf50) double mutants have a shortened life span indistinguishable from daf-16(mgDf50) single mutants. daf-16(mgDf50) is epistatic to skn-1(RNAi).
Whereas daf-2(e1370) results in increased levels of pdp-1 mRNA, daf-16(mgDf50) results in reduced levels of pdp-1 mRNA. daf-2(e1370);daf-16(mgDf50) double mutants have reduced pdp-1 mRNA levels similar to daf-16 single mutants
When RLE-1 expression was disrupted in daf-16::gfp animals by crossing rle-1 mutants into the daf-16::gfp background, the lifespan of daf-16::gfp increased an additional 10%.
When DAF-16::GFP animals were shifted from 20C to 35C, DAF-16::GFP translocated into the nucleus within 3 hours. DAF-16::GFP; apl-1(yn5) animals showed a delayed DAF-16::GFP nuclear translocation compared to DAF-16::GFP animals at 35C.
Mutating daf-16 did not further enhance the sensitivity of shc-1 to oxidative stress. The daf-16shc-1 double mutant exhibited sensitivity to paraquat similar to shc-1 and daf-16 single mutants.
daf-16(mgDf50) results in decreased levels of ftn-1 mRNA, as determined by qRT-PCR. daf-2(m577) alone results in increased levels of ftn-1 mRNA. daf-16(mgDf50);daf-2(m577) double mutants exhibit ftn-1 mRNA expression indistinguishable from daf-16(mgDf50) single mutants suggesting that daf-16(mgDf50) is epistatic to daf-2(m577) with respect to ftn-1 mRNA expression levels
