In 80% of mir-48mir-241; mir-84 triple mutants at the L3 stage, hbl-1::gfp::hbl-1 expression was detected in hyp7. In contrast, hbl-1::gfp::hbl-1 expression was detected in hyp7 in only 11% of wild-type worms at the L3 stage.
NEKL-2 affects endocytosis of LRP-1::GFP from the apical surface of hyp7. The major localization of LRP-1::GFP in hyp7 of living worms changes fromendosomes in the apical region (control RNAi in wild type) to a smeared pattern on the apical surface upon treatment with nekl-2 RNAi.
Expression in the head hypodermis and the excretory cell remains upon fasting, but additional strong expression of far-7::GFP is observed in the hypodermal syncytium (hyp7) covering the body of the animal.
Reduction of HBL-1 by either the mutant allele or 50% hbl-1(RNAi) led to precocious expression of the let-7::gfp in the seam cells and the VPCs and persistence of GFP expression in the hyp7.
Expression of col-19::GFP expression was reduced in the hyp7 cell, but to a lesser extent than controls, suggesting that hbl-1 RNAi suppresses the heterochronic phenotype of mir-48(n4097); mir-84(n4037)
Expression of col-19::GFP expression was reduced in the hyp7 cell, but to a lesser extent than controls, suggesting that hbl-1 RNAi suppresses the heterochronic phenotype of nhl-2(ok818); ain-1(ku322)
"localization of GSP-2 to epidermal junctions depends on APE-1."; "they localized to repetitive punctate structures lining the cell-cell junctions between the two major epidermal syncytia: hyp7 and the lateral seam cells"
Expression of col-19::GFP expression was reduced in the hyp7 cell, but to a lesser extent than controls, suggesting that hbl-1 RNAi suppresses the heterochronic phenotype of cgh-1(ok492); mir-48(n4097); mir-84(n4037)
