Deletion of mir- 57 significantly increases the
nob-1b transcript level. In the wild type background, the transgene arrays, because they contain the mir- 57 promoter region, should result in overexpression of
mir-57 from the genomic copy. As predicted, expression of the reporter gene is significantly reduced by the presence of the binding site in
nob-1b. The reduction in reporter construct expression was not observed in the absence of a functional
mir-57 gene, indicating that
nob-1 is a direct functional target of
mir-57.
