The levels of the cpi-2a transcript in each stage of development were measured and compared with the previously determined transcript levels of C. elegans Ce-cpl-1 and cpz-1. The graph shows the ratio +/- S.D. between cpi-2a or cpl-1 (A) and cpi-2a or cpz-1 (B) levels and those of the constitutively expressed control gene ama-1 (y axis). The ama-1 gene was used as an internal control transcript to allow the relative quantification of cpi-2a, cpl-1, and cpz-1 expression in distinct developmental stages (x axis). The RNA from mixed stage embryos and from synchronized larval and adult populations collected at 2-h intervals was isolated for the preparation of stage-specific cDNA. Stage-specific molting within the life cycle is indicated by a vertical arrow. Each experimental point was repeated at least twice.
