Transfected cells were stained with DAPI (blue) and immunostained with anti-FLAG (red) and anti-a-tubulin (green) antibodies. Merged images are also shown. (B) A C. elegans gonad from the early L4 stage was dissected. Fixed gonad was stained with DAPI (blue) and immunostained with an anti-FIGL-1 antibody (orange). Merged images are also shown. (C) FIGL-1 was detected with an anti-FIGL-1 antibody at several growth stages in the C. elegans wild-type N2 strain. Ten micrograms of lysates were used for each sample. (D) Depletion of FIGL-1 resulted in defective gonad formation and the sterile phenotype. Feeding RNAi method was employed to deplete FIGL-1. Gonads of progeny were dissected, stained with DAPI, and observed. Arrows indicate normal gonads. Arrowheads indicate defective gonads.
