TreatmentThe daf-16(mgDf50) mutants carrying high copies of daf-16a::gfp transgene were maintained at 15 degrees Celsius on RNAi plates for two generations. The plates were then transferred to 20 degrees Celsius for 18 hours. About 15-20 L4 worms were then mounted on slides and visualized by fluorescence microscopy.
