Four kilobases of sequence upstream of each translational start site was fused to GFP to produce the mig-10b::gfp transcriptional fusion construct. An integrated transgene, mpIs1, was created by injection of dpy-20(e1282ts) animals with 100 ug/ml dpy-20(+) DNA and 50 ug/ml of mig-10b::gfp. One of two similar resulting extrachromosomal array strains (mpEx2) was integrated. One hundred L4 dpy-20(e1282ts); mpEx2 animals were irradiated with a gamma source (4000 Rads). | ||
