| Construction_summary | Clone = pTU819. An unstable GFP reporter generated by fusing GFP to a RING-finger domain from the mouse E3 ubiquitin ligase PJA1/C-1. Plasmid vector (TU819) containing Pmec-18::praja::gfp was constructed by first isolating the Praja domain from a mouse cDNA library by PCR using the Praja primers. Nested PCR was then used to generate a DNA fragment fusing the Praja domain to the 5' end of gfp; the PCR also introduced an ATG 5' to the praja::gfp fusion and KpnI and ApaI cloning sites flanking the fragment. The sequence for GFP was taken from the pPD95.75 expression vector. KpnI and ApaI cloning sites were then used to replace gfp with praja::gfp in the pPD95.75 vector. A 0.4-kb PCR fragment containing the mec-18 promoter was then inserted into the HindIII-BamHI sites before praja::gfp. | ||
