| Antigen | Protein | A portion of pgl-3 cDNA (279 bp) corresponding to the PGL-3 region from Ser-448 to Ser-540, which is relatively unique to PGL-3, was PCR amplified using primers 5'-GAGGGATCCAGTTCGTCGGAACCTTCTGC-3' and 5'-TCCCAAGCTTAGCTAGAAAATTGAACAGGTG-3' and cloned into the expression vector pET-28a(+) (Novagen). The 6x His-tagged fusion protein was expressed in Escherichia coli BL21(DE3) by isopropyl thiogalactoside (IPTG) induction and purified using Ni-NTA agarose columns (QIAGEN, Chatsworth, CA) under denaturing conditions. The purified fusion protein was electrophoresed on SDS-polyacrylamide gels, cut from the gels, and injected with Freund's adjuvant into rats. | ||
