A 4-kb region upstream of the predicted ATG start codon of HMR-1B was amplified from a lysate of wild-type worms using the primers HMR1Bp-PstI and HMR1Bp-BamHI. The resulting PCR product was cloned into the GFP expression vector pPD95.69, resulting in an in-frame fusion of the GFP-coding region with the predicted first two codons of HMR-1B. | ||
