| Construction_summary | [unc-17::gfp] translational fusion. A 7832 bp PstI-BamHI unc-17-containing fragment of cosmid ZC416 was inserted into the PstI-BamHI sites in pPD95.75 to make pR14. The GFP sequence was fused in frame to the last exon of unc-17. Upstream sequences of unc-17 were added by inserting a 8755 kb PstI fragment of ZC416 into the PstI site of pR14 to make pR18. pR18 (100 ng/ul) and the wild-type pha-1 plasmid pBX-1 (100 ng/ul) were injected into the germline of pha-1; him-5 hermaphrodites.
