| Construction_summary | Clone = pCF1021. The bi-cistronic rde-1:SL2:sid-1 entry clone was generated using In-Fusion PCR cloning kit (Clonetech) in two steps: first, the 245 bp SL2-specific trans-splice site from the gpd-2/gpd-3 intergenic region was inserted upstream of the start codon of the sid-1 entry vector, then the rde-1 gene was inserted upstream of the SL2 site. The Punc-47 promoter entry construct was a gift from Gunther Hollopeter, University of Utah, Salt Lake City, UT. The expression clone was generated using Gateway LR clonase II Plus (Life Technologies) and inserted into pCFJ150, a Gateway three-fragment compatible destination vector for MosSCI containing a C. briggsae unc-119 rescue fragment and genomic regions flanking the ttTi5605 Mos1 insertion. | ||
