Either 10 ng of total RNA (lanes 1-4) or 3 ng of poly(A) RNA (lanes 5-12) were electrophoresed through 1.2% agarose, 6 M formaldehyde gels and transferred to nitrocellulose filters. The stage-specific RNAs analyzed were: embryo RNA (lanes 1, 5, and 9), L2-dauer RNA (lanes 2, 6, and 10), dauer-L4RNA (lanes 3, 7, and I I ) , and L4-adult RNA (lanes 4,8, and 12). The filters were probed with nick translated plasmids. To detect the total collagen transcripts the probe was pCol-2a (lanes 1-4) used with low stringency conditions. To detect col-1 specific and col-2 specific transcripts the probes were pCol-lb (lanes5 and 6) and pCol-2b (lanes 9-12), respectively, used with high stringency conditions. The hybridized filters were exposed to XAR-5 film with intensifying screens for 22 h (lanes 1-4) or 72 h (lanes 5-12).
