DPY-27 is localized to the nucleus of wild-type XX embryos. The level of DPY-27 is greatly diminished in embryos carrying the null allele dpy-27(y767), thus demonstrating the specificity of the antibody for DPY-27. The ~767 mutation creates an in-frame stop codon at amino acid position 455. The intense staining of dpy-27(y767) embryos with anti-UNC-88 antibodies indicates that reduced staining with anti-DPY-27 antibodies is not caused by a general reduction in protein synthesis or a general increase in protein degradation in these dying embryos. The large reduction in the level of DPY-27 epitope is probably due to the instability of either the peptide or the partially translated mRNA. Scale bar, 20 um.
