An 8 kb sma-3 genomic fragment was obtained from the cosmid R13F6 by digesting with PstI and subcloning into the vector pBLUESCRIPT SK+ (pCS29). This construct rescues sma-3(wk30) mutants. To create GFP fusion constructs, MluI restriction sites were created in the sma-3-coding region, after the start codon (pCS185) or before the stop codon (pCS186) independently, by site-directed mutagenesis. A GFP MluI fragment from pPD118.90 was subcloned into the newly generated MluI sites, forming two sma-3::gfp translational fusion constructs; N-terminal GFP (pCS170) or C-terminal GFP (pCS171). --precise ends. | ||
