Construction_summaryThe marker transgene Pmyo-2::dsRED was used at a concentration of 10 ng/l as a coinjection marker as previously described (Guthrie et al., 2009). The TDP-43 construct was microinjected into N2 at a concentration of 160-180 ng/l as previously described (Kraemer et al., 2003) to produce worms with TDP-43 or green fluorescent protein (GFP) and dsRED fluorescent protein (RFP) transgenes as extrachromosomal arrays. Transgenes were integrated by exposing animals to a dose of 3200-3600 rad gamma rays and integrated strains were backcrossed to N2 at least twice.
