TreatmentSynchronized embryos were hatched overnight in M9 medium to obtain an L1 synchronized population that was seeded onto RNAi plates and grown to adult hermaphrodites (P0). P0 gravid hermaphrodites were used to obtain an L1 synchronized population (F1) that was seeded onto RNAi plates and allowed to grow to adults. Gravid hermaphrodites were transferred to RNAi plates containing Tunicamycin (0 or 2.5 micrograms/ml) for 6 h to lay eggs after which adults were removed. The developmental stages of F2 were analyzed for 5 days at intervals of 24 h and the percentages of L1, L2/L3, L4 and adults were determined.
