WBPicture0000009843DescriptionFigure S3. Patterns of Expression of na/nca and unc-79. (A) Reporter constructs linking the promoters for each channel gene to green fluorescent protein (GFP). For the nca-1 construct (top), the fluorescence pattern exhibited by a typical L3 hermaphrodite is shown. GFP expression is observed in a subset of neurons in the pharyngeal nervous (PN) system, in numerous unidentified nerve ring (NR) interneurons, and in a subset of motor neurons of the ventral nerve cord (VNC). Expression is also observed in a few cell bodies of the dorsorectal and lumbar ganglia (bracket). For the nca-2 construct (bottom), the fluorescence pattern exhibited by a typical L4 hermaphrodite is shown. Expression is observed in a few neurons of the pharyngeal nervous system (PN), in numerous unidentified NR interneurons, in most motor neurons of the VNC, and in neurons and muscle cells in the tail (arrow). Diffuse GFP expression is also observed in vulval muscles (VM) and diffusely in the intestine (INT). Scale bars represent 50 mm. We would like to thank A. Fire (Stanford University) for providing the plasmids pPD95.67 and pPD95.75 used to generate the nca-1 and nca-2 GFP reporter constructs. (B) A reporter construct linking the unc-79 promoter to GFP shows multiple patterns of expression in C. elegans. In adults, the predominant pattern (top left) involves staining of one or two neurons in the head, with processes extending to the snout. Expression was rarely seen in an additional set of paired neurons in the tail (bottom left). In L2 and L3 animals (right), cells of the ventral nerve cord are seen exclusively.
