Construction_summaryThe presynaptic BLINC construct encodes a protein in which the signal sequence of the presynaptic cell adhesion protein NRX-1/neurexin was replaced by the heterologous signal sequence of PAT-3 -integrin followed by the E. coli biotin ligase BirA, it is based on modified GRASP constructs, which had previously been shown to be correctly trafficked to the cell surface (FEINBERG et al. 2008). The N-terminus of the postsynaptic cell adhesion molecule NLG-1/neuroligin was modified such that the endogenous signal peptide was replaced by the PAT-3 -integrin signal sequence followed by two copies of the fifteen amino acid acceptor peptide (AP). NLG-1/neuroligin is a post-synaptic partner of neurexin (reviewed by (CRAIG AND KANG 2007)).
